Viral inosine triphosphatase: A mysterious enzyme with typical activity, but an atypical function

چکیده

Plant viruses typically have highly condensed genomes, yet the plant-pathogenic Cassava brown streak virus, Ugandan cassava and Euphorbia ringspot virus are unusual in encoding an enzyme not found any other “house-cleaning” inosine triphosphatase. Inosine triphosphatases (ITPases) conserved enzymes that occur all kingdoms of life perform a house-cleaning function by hydrolysing noncanonical nucleotide triphosphate to monophosphate. The ITPases encoded been characterized biochemically shown typical ITPase activity. However, their biological role infection has be elucidated. Here we review what is known viral-encoded speculate on potential roles with aim generating greater understanding viruses, group world's most devastating viruses.

Pharmacogenetic significance of inosine triphosphatase.

Inosine triphosphatase (ITPase) is the enzyme that catalyzes the conversion of inosine triphosphate (ITP) and deoxy-inosine triphosphate (dITP) to inosine monophosphate and deoxy-inosine monophosphate, respectively, thereby maintaining low intracellular concentrations of ITP and dITP. Individuals deficient in ITPase activity were first recognized over 30 years ago. For decades, no clinical sign...

Erythrocyte Inosine Triphosphatase Activity Is Decreased in HIV-Seropositive Individuals

BACKGROUND Inosine triphosphatase (ITPase) is encoded by the polymorphic gene ITPA and maintains low intracellular levels of the inosine nucleotides ITP and dITP. The most frequently reported polymorphisms are ITPA c.94C>A (rs 1127354) and ITPA c. 124+21 A>C (rs7270101). Some nucleoside-analogues used in the treatment of HIV-seropositive (HIV+) patients are potential substrates for ITPase. Ther...

A comparative study of inosine triphosphatase activity in fresh erythrocytes and dried blood spots.

Inosine triphosphatase (ITPase) is one of the enzymes involved in thiopurine metabolism. Polymorphisms in the ITPA gene, resulting in a decreased activity of ITPase, are associated with adverse drug reactions during thiopurine therapy. Measuring ITPase activity is therefore of utmost importance. We compared measurement of ITPase activity in fresh erythrocyte lysates and eluates of dried blood s...

Erythrocyte Inosine triphosphatase activity: A potential biomarker for adverse events during combination antiretroviral therapy for HIV

The purine analogues tenofovir and abacavir are precursors of potential substrates for the enzyme Inosine 5'-triphosphate pyrophosphohydrolase (ITPase). Here, we investigated the association of ITPase activity and ITPA genotype with the occurrence of adverse events (AEs) during combination antiretroviral therapy (cART) for human immunodeficiency virus (HIV) infection. In 393 adult HIV-seroposit...

Mysterious but not serious?

1 of 2 DESCRIPTION A previously well 13-year-old boy presented with a 3-month history of right-sided hip pain responding to simple analgesia. Physical examination and blood tests were unremarkable. A pelvic x-ray ( fi gure 1 ) revealed multiple sclerotic lesions in both femora, worse on the asymptomatic left side ( fi gure 2 ) with further lesions in the pelvis. Review of a previous x-ray taken...
